Specificity: Clone G28.5 recognizes an epitope on human CD40.
Endotoxin Level: <0.5 EU/mg as determined by the LAL method.
Purity: ≥98% monomer by analytical SEC and >95% by SDS Page.
Preparation: Functional grade preclinical antibodies are manufactured in an animal free facility using in vitro cell culture techniques and are purified by a multi-step process including the use of protein A or G to assure extremely low levels of endotoxins, leachable protein A or aggregates.
Pathogen Testing: To protect mouse colonies from infection by pathogens and to assure that experimental preclinical data is not affected by such pathogens, all of Leinco Purified Functional PLATINUM™ antibodies are tested and guaranteed to be negative for all pathogens in the IDEXX IMPACT I Mouse Profile.
Storage: Functional grade preclinical antibodies may be stored sterile as received at 2-8C for up to one month. For longer term storage, aseptically aliquot in working volumes without diluting and store at -80C. Avoid Repeated Freeze Thaw Cycles.
Target Information
The CD40 antigen is a single chain glycoprotein that is known to be a member of the tumor necrosis factor/nerve growth factor superfamily and shows a significant hom*ology to the Hodgkin's disease associated antigen, CD30. CD40 is present on all B cells except plasma cells and is also found on some epithelial cells, carcinomas and lymphoid dendritic cells. CD40 has been found to be essential in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. AT-hook transcription factor AKNA is reported to coordinately regulate the expression of CD40, which may be important for hom*otypic cell interactions. Adaptor protein TNFR2 interacts with CD40 and serves as a mediator of the signal transduction. The interaction of CD40 and its ligand is found to be necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis. Two alternatively spliced transcript variants of CD40 encoding distinct isoforms have been reported. Diseases associated with CD40 dysfunction include Type 3 Hyper-Igm immunodeficiency and CD40 ligand deficiency.
